You will need both prior to submission of application.
Both need to be at UKBA approved centres

English test you will need to submit pass certificate number on application form
Tb screening certificate submit with supporting documents

Believe you want to use link below to apply for visa
    https://www.gov.uk/uk-family-visa/partner-spouse

The visa you want will be appendix FM partner

This is settlement visa for your wife as you are applying under appendix FM rules.

Worth making yourself aware of evidence requirements under appendix FM

follow these steps and you will get there, the biometrics is last
1 start application and gather all required documents
2 do English test and get TB screening done without these you cannot progress your application
3 complete application pay NHS surcharge and submit application
4.upload documents or take with you to biometric centre where they will scan them for you
    5 complete biometrics
